As of now you can easily prevent spammy and bots’ massages on your networks using CAPTCHA option.
It is available both for Ning 2.0 and Ning 3.0 platforms.
You can enable CAPTCHA in General Settings of your Network. Simply go to Social Site Manager section, if you are on Ning 3.0 or to Settings Network Details if you are on Ning 2.0 .

Even if you disable it, your new member will see CAPTCHA once when they're on registration page.

As another example I added @yandex.com a few days ago and this afternoon I get another one of those spammers getting through. They are in my 'awaiting approval' list at the moment if you want to look. Does this mean that using domain blocking does not work?

It seems that for some reason domain blockage hasn't worked out for that user specifically, as when I have tried to sign up before by using the domains from the blocked list, there always was the error message.
Sorry for any inconveniences, we have filed the report to our tech team to find out who this person has gone around the blockage.
Please don't approve that person or remove him/her from the network, it's necessary to keep this profile on the list to run several tests.
